Multiple Antigenic Peptides (MAPs)
MAPs stands are Multiple Antigenic Peptide, is a branched peptide at which linear peptide chains are linked at their C-terminus via polylysine core, there by increasing the size of whole molecule. Genesis Biotech Inc. provides several choices for MAP such as 4 branches asymmetric or Symmetric, and 8 branches asymmetric or symmetric. Therefore, it can enhance antigen by increasing peptideˇ¦s molecular weight. If we choose MAP as antigen, it is unnecessary to conjugate carrier protein..
|
MAP is simply an alternative to KLH conjugation. This is done to eliminate the coupling of peptides to KLH. It is recommended only for short peptides located either internally within a protein or at the N-terminus.KLH conjugation is recommended for a peptide at any location in a protein and when there is a cysteine residue within the peptide sequence.
It seems that, however, conformation of peptides on MAP is less flexible, and antibodies obtained by MAP often recognize protein less often than by conventional KLH conjugation. In addition, there is no free peptide produced when making MAP, making it difficult to remove polylysine core directed antibodies. Purification of MAP by HPLC is difficult, and MAP is provided without mass verification due do its heterogeneity and large molecular size.

Application
Multiple antigen peptide application is one potent
way to produce high-titer anti-peptide antibodies and synthetic
peptide vaccines. This system utilizes the a- and e-amino groups of
lysine to form a backbone to which multiple peptidechains can be
attached. Depending on the number of lysine tiers, different numbers
of peptide branches can be synthesized. This eliminates the need to
conjugate the antigen to a protein carrier.
